Levosimendan protects against ischemia – reperfusion injury in the human heart muscle. A pilot study

چکیده

Abstract The consequences of myocardial infarction (MI) are an increasing problem worldwide. Despite spectacular progress in the invasive treatment ischemic heart disease, ability to limit ischemia-reperfusion (I/R) injury remains largely unrealized. Recent studies have shown that stimulation opioid receptors may confer a cardioprotective effect against I/R injury. Levosimendan, inodilator, is indicated for short-term acutely decompensated failure. We tested hypothesis levosimendan provide cardioprotection opioid-like mechanism human myocardium.

Levosimendan protects human hepatocytes from ischemia-reperfusion injury

BACKGROUND Ischemia-reperfusion injury (IRI) is a major challenge in liver transplantation. The mitochondrial pathway plays a pivotal role in hepatic IRI. Levosimendan, a calcium channel sensitizer, was shown to attenuate apoptosis after IRI in animal livers. The aim of this study was to investigate the effect of levosimendan on apoptosis in human hepatocytes. METHODS Primary human hepatocyte...

Cardiac mTOR protects the heart against ischemia-reperfusion injury.

Cardiac mammalian target of rapamycin (mTOR) is necessary and sufficient to prevent cardiac dysfunction in pathological hypertrophy. However, the role of cardiac mTOR in heart failure after ischemic injury remains undefined. To address this question, we used transgenic (Tg) mice with cardiac-specific overexpression of mTOR (mTOR-Tg mice) to study ischemia-reperfusion (I/R) injury in two animal ...

Inhibiting mitochondrial fission protects the heart against ischemia/reperfusion injury.

BACKGROUND Whether alterations in mitochondrial morphology affect the susceptibility of the heart to ischemia/reperfusion injury is unknown. We hypothesized that modulating mitochondrial morphology protects the heart against ischemia/reperfusion injury. METHODS AND RESULTS In response to ischemia, mitochondria in HL-1 cells (a cardiac-derived cell line) undergo fragmentation, a process that i...

RhoA protects the mouse heart against ischemia/reperfusion injury.

The small GTPase RhoA serves as a nodal point for signaling through hormones and mechanical stretch. However, the role of RhoA signaling in cardiac pathophysiology is poorly understood. To address this issue, we generated mice with cardiomyocyte-specific conditional expression of low levels of activated RhoA (CA-RhoA mice) and demonstrated that they exhibited no overt cardiomyopathy. When chall...

Apelin protects heart against ischemia/reperfusion injury in rat.

Apelin, the endogenous ligand of the G protein-coupled APJ receptor, is a peptide mediator with emerging regulatory actions in the heart. We aimed to determine whether the endogenous apelin/APJ system is an intrinsic protective pathway in ischemic/reperfusion injury. A Langendorff model of perfused isolated rat hearts and primary cultured myocardial cells from neonatal rats were used. Cardiac f...
